The enzyme typically attaches <a href=\"https:\/\/en.wikipedia.org\/wiki\/Fatty_acid\">fatty acid molecules<\/a> to the infamously suspicious <a href=\"https:\/\/www.sciencealert.com\/researchers-have-a-new-theory-about-what-causes-alzheimers-and-its-not-plaque\">beta-amyloid<\/a> and <a href=\"https:\/\/www.sciencealert.com\/scientists-have-uncovered-the-atomic-structure-of-a-key-alzheimer-s-protein-for-the-first-time\">tau protein<\/a> clumps, though in individuals with <a href=\"https:\/\/www.aginganddisease.org\/EN\/10.14336\/AD.2023.0814\">brain insulin resistance (BIR)-dependent deterioration<\/a> the process can get out of control.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;In this new study, we showed that in the early stages of Alzheimer&#8217;s molecular changes resembling a scenario of brain insulin resistance cause an increase of S-acyltransferase enzyme levels,&#8221; <a href=\"https:\/\/www.eurekalert.org\/news-releases\/1065362\">says<\/a> neuroscientist Salvatore Fusco, explaining how these enzymes then go on to alter cognitive function and the abnormal accumulations of protein clumps.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>Beta-amyloid and tau are naturally occurring proteins that increase in Alzheimer&#8217;s brains as the disease progresses.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>Understandably, this has made the protein clumps a huge focus in Alzheimer&#8217;s research. Yet, in a seeming contradiction, laboratory studies suggest the <a href=\"https:\/\/www.sciencealert.com\/we-might-been-wrong-what-kills-brain-cells-alzheimer-s-disease-beta-amyloid-tau-protein-app\">protein clumps do not directly damage brain cells<\/a>. (<a href=\"https:\/\/doi.org\/10.1126\/sciadv.adp5059\">Nirmalraj et al.,<span> <\/span><em>Science Advances<\/em>, 2024<\/a><span>)<\/span><\/figcaption><\/figure>\n<p>So Natale and team disabled the function of the S-acyltransferase enzyme in mice genetically modified to have a mouse version of Alzheimer&#8217;s disease.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>Alzheimer&#8217;s symptoms reduced whether the enzyme was turned off genetically or counteracted with an agent delivered through a nasal spray. Each treatment also seemed to slow the neurodegeneration and extend the rodent&#8217;s lifespans.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>The team did not detect any effects in normal mice given the same treatment.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>The active agent in the nasal spray, 2-bromopalmitate, carries a high risk of <a href=\"https:\/\/doi.org\/10.1016\/j.cbpa.2021.07.002\">interfering with a number of processes<\/a>, making it unsafe for testing in humans. But the team is hopeful an alternative could be identified, now they have a specific target.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>More studies will be needed before researchers can determine if this is a safe tactic. But with a new dementia diagnosis now being made <a href=\"https:\/\/www.sciencealert.com\/alzheimers-might-not-actually-be-a-brain-disease-expert-reveals\">every three seconds<\/a>, and no cure, better treatments are urgently needed.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;New approaches \u2013 potentially translatable to human therapies \u2013 will be tested, including &#8216;genetic patches&#8217; or engineered proteins that can interfere with [S-acyltransferase] enzyme activity,&#8221; <a href=\"https:\/\/www.eurekalert.org\/news-releases\/1065362\">says<\/a> neuroscientist Claudio Grassi.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>The team&#8217;s findings parallel another recent study that also suggests the problematic beta-alpha protein clumps can be both involved and somehow not involved in damaging brain tissues <a href=\"https:\/\/www.sciencealert.com\/newly-identified-proteins-may-explain-devastating-effects-of-alzheimers\">depending on the other molecules present<\/a> with them.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;To date no therapeutic interventions targeting\u2026 [S-acyltransferase] have been attempted in Alzheimer&#8217;s disease.
