Scientists Say That&#8217;s The Point. : ScienceAlert"},"content":{"rendered":"<p> <script async src=\"https:\/\/pagead2.googlesyndication.com\/pagead\/js\/adsbygoogle.js?client=ca-pub-3711241968723425\"\r\n     crossorigin=\"anonymous\"><\/script>\r\n<ins class=\"adsbygoogle\"\r\n     style=\"display:block\"\r\n     data-ad-format=\"fluid\"\r\n     data-ad-layout-key=\"-fb+5w+4e-db+86\"\r\n     data-ad-client=\"ca-pub-3711241968723425\"\r\n     data-ad-slot=\"7910942971\"><\/ins>\r\n<script>\r\n     (adsbygoogle = window.adsbygoogle || []).push({});\r\n<\/script><br \/>\n<\/p>\n<div>\n<p>Decreases in brain volume are used as <a href=\"https:\/\/doi.org\/10.1016\/j.jalz.2018.02.018\">an indicator of Alzheimer&#8217;s disease<\/a>, yet the latest class of  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"144855\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a> treatments cause the brain to shrink even further.<\/p>\n<p>A new study now suggests this might paradoxically be a good thing.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>Neurosurgeon Christopher Belder from University College London (UCL) and colleagues found the loss of brain volume in patients undergoing new immunotherapy treatments is likely caused by a successful decrease of the suspicious protein clumps clogging Alzheimer&#8217;s brains, rather than the drug destroying tissues.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;Amyloid immunotherapy has consistently shown an increase in brain volume loss \u2013 leading to concerns in the media and medical literature that these drugs could be causing unrecognized toxicity to the brains of treated patients,&#8221; <a href=\"https:\/\/www.ucl.ac.uk\/news\/2024\/nov\/brain-shrinkage-associated-alzheimers-therapies-shows-effectiveness-rather-harm\">explains<\/a> UCL neurologist Nick Fox.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;However, based on the available data, we believe that this excess volume change is an anticipated consequence of the removal of pathologic amyloid plaques from the brain of patients with Alzheimer&#8217;s disease.&#8221;<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>Belder, Fox, and colleagues analyzed data from twelve different Alzheimer&#8217;s treatment trials targeting the naturally-occurring beta-amyloid proteins which clump excessively in patients with Alzheimer&#8217;s.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>While there is now <a href=\"https:\/\/www.sciencealert.com\/reigning-theory-of-alzheimer-s-may-be-based-on-fraudulent-images-per-new-report\">some controversy<\/a> around the protein clumps&#8217; role in Alzheimer&#8217;s, several <a href=\"https:\/\/www.sciencealert.com\/nasal-spray-may-slow-alzheimers-disease-study-in-mice-suggests\">recent studies<\/a> suggest any harm caused by the plaques may depend on surrounding molecules which share the protein&#8217;s environment.<\/p>\n<figure id=\"attachment_142437\" aria-describedby=\"caption-attachment-142437\" style=\"width: 1204px\" class=\"wp-caption alignnone\"><img decoding=\"async\" src=\"https:\/\/www.sciencealert.com\/images\/2024\/11\/AmyloidPlaquesAmongNeurons_R_U_642.jpg\" alt=\"Clumps of yellow forming among purple neurons\" width=\"1204\" height=\"938\" class=\"size-full wp-image-142437\" srcset=\"https:\/\/www.sciencealert.com\/images\/2024\/11\/AmyloidPlaquesAmongNeurons_R_U_642.jpg 1204w, https:\/\/www.sciencealert.com\/images\/2024\/11\/AmyloidPlaquesAmongNeurons_R_U_642-642x500.jpg 642w, https:\/\/www.sciencealert.com\/images\/2024\/11\/AmyloidPlaquesAmongNeurons_R_U_642-533x415.jpg 533w, https:\/\/www.sciencealert.com\/images\/2024\/11\/AmyloidPlaquesAmongNeurons_R_U_642-768x598.jpg 768w, https:\/\/www.sciencealert.com\/images\/2024\/11\/AmyloidPlaquesAmongNeurons_R_U_642-600x467.jpg 600w\" sizes=\"auto, (max-width: 1204px) 100vw, 1204px\" loading=\"lazy\"\/><figcaption id=\"caption-attachment-142437\" class=\"wp-caption-text\">Illustration of amyloid plaques (yellow) among neurons and other molecules. What&#8217;s more, the level of volume loss reflected changes in the levels of beta-amyloid.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;The volume occupied by beta-amyloid plaques in the brains of people with Alzheimer&#8217;s disease is not trivial (around 6 percent of cortex according to post-mortem studies),&#8221; Belder and team <a href=\"https:\/\/doi.org\/10.1016\/S1474-4422(24)00335-1\">explain<\/a>.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;The extent of excess volume change seen in treated patients is considerably lower than this volume occupied by plaques.&#8221;<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>Their proposed explanation is still incomplete, Belder and team caution, noting possible shifts in brain fluid makes multiple factors likely.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;Given that some of these anti-amyloid treatments are now in clinical use and others are in or entering  <a href=\"https:\/\/www.sciencealert.com\/clinical-trials\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73032\" data-postid=\"144855\" rel=\"nofollow noopener\" target=\"_self\">clinical trials<\/a>, it is vital to understand whether these volume changes are a signal of harm,&#8221; the researchers <a href=\"https:\/\/doi.org\/10.1016\/S1474-4422(24)00335-1\">warn<\/a>.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;There are many unanswered questions, including the long term trajectory of volume changes.
