Yet the condition commonly affects other neurological functions as well, impacting mood and causing a decline in cognition.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>A drug that blocks a key receptor linked with blood pressure has shown promise recovering memory in <a href=\"https:\/\/www.aginganddisease.org\/EN\/10.14336\/AD.2023.0928\">models of vascular dementia<\/a>, inspiring researchers from the University of Arizona to test the treatment on mice with <span>Parkinson&#8217;s<\/span>-like symptoms.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>Called PNA5, the short peptide is an attractive choice of medication given its affinity for its target and ability to be broken down safely in the body. The researchers analyzed the memories and tissue samples of the mice to determine whether changes could be attributed to the drug.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>The team concluded doses of PNA5 reversed some signs of cognitive decline in the mice, while a reduction in the activity of immune cells called <a href=\"https:\/\/en.wikipedia.org\/wiki\/Microglia\">microglia<\/a>, which are thought to contribute to cognitive decline when they get pushed into overdrive, was observed in their hippocampus.<\/p>\n<figure id=\"attachment_145465\" aria-describedby=\"caption-attachment-145465\" style=\"width: 642px\" class=\"wp-caption alignnone\"><img decoding=\"async\" src=\"https:\/\/www.sciencealert.com\/images\/2024\/12\/MouseTasks.jpg\" alt=\"Mouse tasks\" width=\"642\" height=\"242\" class=\"wp-image-145465 size-full\" srcset=\"https:\/\/www.sciencealert.com\/images\/2024\/12\/MouseTasks.jpg 642w, https:\/\/www.sciencealert.com\/images\/2024\/12\/MouseTasks-600x226.jpg 600w\" sizes=\"auto, (max-width: 642px) 100vw, 642px\" loading=\"lazy\"\/><figcaption id=\"caption-attachment-145465\" class=\"wp-caption-text\">The researchers put mice through challenges to test their cognition. (Bernard et al., <em>Experimental Neurology<\/em>, 2024)<\/figcaption><\/figure>\n<p>&#8220;With PNA5, we&#8217;re targeting cognitive symptoms but, in particular, we&#8217;re trying to prevent further degeneration from occurring,&#8221; <a href=\"https:\/\/healthsciences.arizona.edu\/news\/releases\/study-identifies-potential-new-drug-parkinsons-related-cognitive-decline-dementia\">says<\/a> neurobiologist Kelsey Bernard. &#8220;By going down the protective route, we can hopefully prevent cognitive decline from continuing.&#8221;<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>The treatment in the mice was enough to improve <a href=\"https:\/\/en.wikipedia.org\/wiki\/Recognition_memory\">recognition memory<\/a> and <a href=\"https:\/\/en.wikipedia.org\/wiki\/Spatial_memory\">spatial working memory<\/a>, the researchers found. The loss of brain cells in their hippocampus was also slowed when PNA5 was introduced.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>By calming the microglia cells down to a more normal state, it&#8217;s thought that PNA5 can reduce inflammation in the brain, and stop some of the havoc that&#8217;s caused by the body&#8217;s out-of-control defense system.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;Normally, microglia are looking for things like  <a href=\"https:\/\/www.sciencealert.com\/virus\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73053\" data-postid=\"145462\" rel=\"nofollow noopener\" target=\"_self\">viruses<\/a> or injury and secreting substances that block off the damage,&#8221; <a href=\"https:\/\/healthsciences.arizona.edu\/news\/releases\/study-identifies-potential-new-drug-parkinsons-related-cognitive-decline-dementia\">says<\/a> Bernard.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>&#8220;In Parkinson&#8217;s disease, when they&#8217;re constantly activated, microglia can propagate further damage to the surrounding tissue. That&#8217;s what we see in Parkinson&#8217;s brains, particularly in regions associated with cognitive decline.&#8221;<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>PNA5 is produced in the lab by building on the vasoconstricting hormone <a href=\"https:\/\/en.wikipedia.org\/wiki\/Angiotensin\">angiotensin<\/a>, and researchers have already made progress in getting it to penetrate into the brain and last for longer.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>While these are promising results, the drug&#8217;s safety and efficacy needs to be demonstrated in humans. There&#8217;s still more work to be done in understanding how PNA5 impacts cells in our own brain.<\/p>\n<p><!-- START single\/mrec --><br \/>\n<!-- END single\/mrec --><\/p>\n<p>While some the debilitating symptoms of Parkinson&#8217;s can be well managed with the right care, there&#8217;s nothing yet that can be done about the dementia aspect of the disease.
